| 12:40–1:00 pm | **Integrated Bioanalytical Strategies for PK, Biomarker, and Immunogenicity Assessments Across Therapeutic Modalities Supporting Regulatory Approval**, Meina Liang, Ph.D. Pharmacokinetic (PK), biomarker, and immunogenicity assessments are critical to the successful development and regulatory approval of new therapeutics. While regulatory expectations are relatively well established for mature modalities such as small molecules and antibodies, expectations for emerging and novel drug modalities continue to evolve. This presentation will discuss key strategies and considerations for PK, biomarker, and immunogenicity assessments across drug development stages and therapeutic modalities, with a focus on supporting regulatory submissions. Selected case examples will illustrate practical approaches and modality-specific considerations. |
| 1:00–1:20 pm | **Immunogenicity Characterization Tier Analysis for Biotherapeutics: Neutralizing Anti-Drug Antibody and Domain Specificity Assays**, Inna Vainshtein, Ph.D. Immunogenicity assessment is a critical component of biotherapeutic development, as anti-drug antibodies (ADAs) may affect pharmacokinetics (PK), therapeutic efficacy, and patient safety. Regulatory expectations for immunogenicity bioanalytical methods emphasize assay robustness, sensitivity, and drug tolerance to support reliable clinical interpretation. While ADA screening, confirmatory, and titer assays are relatively mature methodologies, characterization assays, including neutralizing antibody (NAb) and domain specificity assays, are often more complex to develop, optimize, and validate. This presentation will discuss the immunogenicity characterization tier, with a focus on regulatory expectations, current challenges, and solutions for NAb and domain specificity assessments. |

This will include ADA strategy for discovery bioanalysis, GLP-Tox, and Clinical Phase 1-3, and will include ADA/NAb examples. |
| 1:40–2:00 pm | **ADC Bioanalysis: Navigating Complex PK and Biotransformation in Drug Development**, Jintang He, Ph.D. This presentation explores bioanalytical strategies for assessing the pharmacokinetics (PK) and biotransformation of antibody-drug conjugates (ADCs), covering conventional assay methodologies, key bioanalytical challenges, and the new opportunities offered by emerging technologies. |
| 2:00–2:20 pm | **Bridging Biology Across Populations: Biomarkers for Global Evidence Generation**, Preeti Paliwal, Ph.D. This presentation explores how biomarker evidence can demonstrate shared disease biology across patient populations, strengthen confidence in cross-regional evidence, and inform global clinical development and regulatory decision-making. |

| 2:45–3:00 pm | **Challenges and Strategies for Clinical Dose Optimization**, Lorin Roskos, Ph.D. For non-oncology indications, randomized, parallel-group Phase 2 studies with efficacy and pharmacodynamic biomarker endpoints have long been the standard approach for selecting the dosing regimen for Phase 3. These studies typically include a suboptimal dose to characterize the exposure–response relationship. In oncology, the traditional approach of identifying a maximum tolerated dose (MTD) from Phase 1 dose-escalation cohorts can be poorly suited to products whose efficacy exposure–response relationship plateaus below the MTD. FDA's Project Optimus has shifted expectations toward comparison of two or more doses before registrational trials, and dose finding is now often conducted in Phase 1 expansion cohorts. The central challenge in oncology is to evaluate doses that may improve safety without using a suboptimal dose that could worsen survival outcomes. For both oncology and non-oncology indications, biomarker strategies can bracket the low and high ends of the dose range under study. Exposure–response and exposure–safety analyses can then be used to quantitatively predict the optimal dose for registrational trials. These approaches can shorten timelines, reduce cost, and increase the probability of success. |
